His tag is a short peptide composed of six histidines (His), which can be inserted into the N-terminal or C-terminal of the target protein, and then separated and purified by the affinity between His and nickel column medium. Because of its low molecular weight and easy separation and purification, His tag is one of the most widely used fusion tags at present.
